Genes and environment both participates in development of cerebral cortex. Anterior part of neural plate in embryo begins the process of cerebral cortex formation. Cavity inside the neural tube (forms by folding of neural plate), develops a ventricular system. Anterior part of the neural plate known as prosencephalon give rise to cerebral cortex.

Ventricular zone contain progenitor cells that forms glial cells and neurons. Progenitor cells divide symmetrically to form glial fibers. But few divide asymmetrically to form two cells. Out of these 2 cells, one cell differentiate in ependymal cell or glial cell. And one cell migrates along the radial glial fibers.

The first pyramidal neurons is generated via migration out from the ventricular zone and subventricular zone, along with reelin producing Cajal–Retzius neurons(reelin producing cells), from preplate.

After this neurons in the middle of the preplate divides to form marginal zone. Marginal zone becomes the first layer of the neocortex, and forms the middle layer known as cortical plate.
