What is the importance of methylation in the activity of restriction endonucleases? restriction enzyme recognizes a unique sequence of bases and hydrolyzes the phosphodiester bond between two nucleotides. When these nucleotides are methylated, the restriction enzyme will not work. What is the importance of "sticky ends" in molecular biology? ends are the overhangs from restriction enzymes that do not cut the ends blunt. The overhanging strand will match up with the complementary single strand from the other side of the cut. This is very in molecular biology because it allows for DNA from different sources to be combined. It doesn't latter if the overhanging ends came from the same DNA or not. As long as both pieces were cut with the restriction enzyme, the pieces can be combined and sealed with DNA ligase.

Restriction enzymes as we know are ezymes that recognizes a species base sequence in a DNA strand, cuts it into fragments known as restriction fragments and then another enzyme as ligase joins them together with another DNA strand sequence. By not having a functional restricted enzymes achieved by methylation would result in an identical DNA sequence and such state is necessary for regrowth process and drug development such as antibiotics selection.
